Conteúdo: Abstract: Cheese ripening is a complex microbial process marked by significant shifts in microbial composition. Considering that propidium monoazide (PMA) depletes DNA from nonviable cells, we hypothesized that PMA treatment of cheese samples could affect the microbiota characterization of 60-d-ripened raw goat curd cheese by 16S rRNA metabarcoding sequencing. After ripening, PMA-treated and nontreated (control) samples from the same cheese units were processed for DNA extraction, library preparation, and 16S rRNA metabarcoding sequencing on an Illumina MiSeq platform. Downstream bioinformatic analyses for microbial diversity assessment were performed using QIIME 2 and the phyloseq package in R. Statistical analyses included permutational multivariate analysis of variance (PERMANOVA), Wilcoxon tests, and linear discriminant analysis effect size (LEfSe). No significant differences were observed in either α or β diversity metrics between PMA-treated and nontreated samples. However, PMA treatment significantly reduced the abundance of farm environment–associated Dickeya and Pectobacteriaceae taxa in cheese samples, thus improving the accuracy of determining the cheese microbial structure using next-generation sequencing technologies. Further longitudinal studies focusing on different sampling periods during ripening, as well as other cheese types, may shed light on the potential benefits of using PMA for improving the accuracy of cheese microbial community characterization by next-generation sequencing. Summary: This study evaluated whether propidium monoazide (PMA) treatment affects the characterization of microbial communities in 60-day-ripened raw goat cheese using 16S ribosomal RNA (rRNA) metabarcoding. The PMA selectively depletes DNA from nonviable cells, potentially providing a more accurate representation of living microorganisms. Paired samples (PMA-treated and nontreated controls) from the same cheese units were analyzed using Illumina MiSeq sequencing, with downstream analyses performed in QIIME 2 and R. Results showed no significant differences in alpha or beta diversity metrics between treatments, indicating that PMA did not alter the overall microbial community structure. However, PMA treatment significantly reduced the abundance of farm environment-associated taxa, specifically Dickeya and Pectobacteriaceae, suggesting these organisms were predominantly nonviable with residual DNA persisting in the cheese matrix. This demonstrates that PMA pretreatment improves the accuracy of cheese microbiome characterization by filtering dead cell DNA from environmental contaminants, providing clearer insights into viable microbial communities during cheese ripening.</description>

Conteúdo: Abstract: The recovery of underutilized fish biomass through enzymatic hydrolysis is an alternative that could contribute to alleviating the problem of malnutrition in developing countries. In this study, a proteolytic enzyme was used to catalyze the hydrolytic cleavage of peptide bonds to produce a fish protein concentrate from shark meat, an underutilized marine species, under conditions of limited hydrolysis. After the enzymatic reaction, the protein was recovered through separation and dehydration. The resulting product was a stable and edible powder that could be utilized as a protein supplement in human nutrition. The final product exhibited high protein (&gt; 82%) and low-fat concentrations (&lt; 0.9%). Its amino acid profile was comparable to that of the original shark muscle, indicating that the nutritional balance was retained. Additionally, the physical characteristics of the spraydried hydrolysates produced from limited hydrolysis of shark meat using the bacterial enzyme Alcalase, suggest potential applications as a protein supplement in protein-rich carbonated beverages.</description>

Conteúdo: [Uso de tampão intravaginal humano embebido com progesterona humana para indução e sincronização de estro em ovelhas Santa Inês]. Resumo: Este estudo avaliou a eficiência de três protocolos de indução de estro síncronizado em ovelhas da raça Santa Inês. Vinte e quatro ovelhas adultas foram equitativamente distribuídas em três grupos, de acordo com ordem de parto, peso corporal (kg) e escore da condição corporal. As ovelhas receberam implante vaginal de progesterona natural por seis dias mais 37,5 g de d-cloprostenol laterovulvar e 300UI de eCG i.m., 24 horas antes da remoção do dispositivo. Ovelhas controle receberam CIDR330mg de progesterona, e as demais receberam dispositivo absorvente intravaginal humano, tamanho mini, embebido com 200 (OB200) ou 400mg (OB400) de progesterona. Coletas de sangue foram feitas nos momentos D0 (antes da inserção dos dispositivos), D0+6h e diariamente, até um dia após retirada do dispositivo (D7). A progesterona (ng/mL) foi semelhante (P&gt;0,05) em todos os tratamentos ao longo do período experimental, exceto no dia da remoção do dispositivo, quando as ovelhas controle (2,5±0,3) tiveram progesterona superior (P&lt;0,05) às ovelhas OB200 (0,6±0,1) e OB400 (1,2±0,4). O percentual de animais em estro (100,0, 62,5 e 100,0%) foi semelhante (P&gt;0,05) e o intervalo para o estro (46,3±3,9a, 26,4±4,5b e 31,2±5,8a,b) foi diferente (P&lt;0,05) entre os animais dos grupos controle, OB200 e OB400, respectivamente. A taxa de concepção das ovelhas foi de 50,0% (4/8) para cada grupo (P&gt;0,05). Dispositivos humanos e fonte de progesterona podem ser usados para induzir o estro sincronizado em ovelhas Santa Inês.</description>

Conteúdo: Abstract: Chemical composition of some potential forages species, natives from Caatinga region, were evaluated. Samples of Macroptilium heterophyllum, Stylosanthes humilis, Rhynchosia mÃ­nima, Desmodium tortuosum Sw. Dc, Merremia aegyptia, Mimosa tenuiflora Wild, Bauhinia cheilantha and as well Macroptilium lathyroides, Caesalpinia pyramidalis and Mimosa tenuiflora hays were collected in Rio Grande do Norte Stated, during 2011 rainy season. The analyses: dry matter (DM), crude protein (CP) mineral matter (MM) ether extractÂ (EE) neutral detergent fiber (NDF), acid detergent fiber (ADF), lignin (LIG), insoluble neutral detergent nitrogen, (INDN) insoluble acid detergent nitrogen, (ADIN), total phenol (TF) and total tannin (TT) were done at Embrapa Caprinos e Ovinos in CearÃ¡ State. Plants analyzed, as expected, for tropical species, exhibited high level of cell wall constituents, high lignifications rate and revealed substantial presence of anti nutritional compounds. However, regardless of this data, the main problem, for grazing animals, is due to its xerophytes characteristics. Most of the shrubs and trees are deciduous, losing its leaves during the dry season. In addition, herbaceous presents a very rapid lifetime cycle, germinating and senescing during the brief wet season. [Composición química y potencial forrajero de especies de la región de Caatinga]. Resumen: Se evaluó la composición química de algunas especies forrajeras potenciales nativas de la región de Caatinga. Muestras de Macroptilium heterophyllum, Stylosanthes humilis, Rhynchosia minima, Desmodium tortuosum Sw. Dc, Merremia aegyptia, Mimosa tenuiflora Wild, Bauhinia cheilantha y así como de heno de Macroptilium lathyroides, Caesalpinia pyramidalis y Mimosa tenuiflora se colectaron en Rio Grande do Norte durante la temporada de lluvias de 2011. Las muestras fueron analizadas para determinar su contenido de materia seca (MS), proteína cruda (PC), minerales (MM), extracto etéreo (EE), fibra neutro detergente (FND), fibra detergente ácido (FDA), lignina (LIG), nitrógeno insoluble detergente neutro (INDN), nitrógeno insoluble detergente ácido (ADIN), fenoles totales (FT) y taninos totales (TT). Como se esperaba, para las especies tropicales, las plantas exhibieron alto nivel de componentes de la pared celular, alta tasa de lignificación y reveló la presencia sustancial de compuestos antinutricionales. Sin embargo, el principal problema para los animales de pastoreo se debe a sus características Xerófitas. La mayoría de los arbustos y los árboles son de hoja caduca, perdiendo sus hojas durante la estación seca. Además, las herbáceas presentan un ciclo de vida muy rápido, germinando y llegando a senescencia durante la breve temporada de lluvias</description>
