dc.descriptione aim was to understand the performance of testicular thermal stress on the morphology, compaction and integrity of DNA in the different phases of the spermatogenic cycle in buffaloes, using chromomycin A3 and acridine orange respectively. Five adult bulls undergoing testicular insulation for 48 hours and semen collected once every seven days, divided into pre and post testicular insulation. e post-insulation testicular temperature increased 3.2ºC (p<0.05) in relation to the control. ere was an increase (p<0.05) in the deprotamination of the spermatic chromatin in the seventh week a#er insulation compared to the control (11.0 ±3.1 and 3.0 ±2.8), but there was no difference in DNA fragmentation a#er insulation. However, head defects showed an increase (p<0.05) in the seventh week in relation to the control (0.7 ±0.2 and 0.13 ±0.16). erefore, the increase in testicular temperature in buffaloes had a negative influence on spermatogonia and primary spermatocytes on chromatin protamination.
