Description: This study aimed to evaluate the meiotic arrest in oocytes using cilostamide, a potent inhibitor of phosphodiesterase type 3, in the maturation medium used for the in vitro embryos production (IVP). For that, 908 bovine oocytes recovered from slaughter house ovaries, were submitted to in vitro maturation for 8 and 24 h. Treatments were: 1) Control (C), culture in maturation medium; 2) Inhibition no hormone (INH), maturation medium no FSH plus cilostamide; 3) Inhibition with hormone (IWH), maturation medium with hormones plus cilostamide; 4) Inhibition with double hormone (IW2H), maturation medium with double of hormone concentration plus cilostamide. After each culture period, oocytes were stained to assess the stage of meiosis. Data were analyzed by χ2 test. Cilostamide completely inhibited resumption of meiosis in the presence or absence of FSH for 8 h, however an increase in FSH concentration (IW2H), overcame its inhibitory effect and the majority of the oocytes resumed meiosis. After 24 h of culture, 95.1% of the oocytes from the C group were mature (MII), while those cultured in presence of cilostamide remained blocked (VG). However, oocytes from IW2H group that had resumed meiosis, at 24 h, showed a delay in meiosis progression with only 79.3% in MII. These results suggest that the meiotic arrest can be successfully attained with cilostamide in maturation medium for 24 hours and can be used as an alternative to improve the competence of oocyte used for the IVP.
