Descripción:	The nematodes of the genus Meloidogyne spp. are distributed worldwide and causes damage in several crops of agronomic importance. This work aims to validate genes involved in metabolism of M. incognita via RNAi. Fragments of Isocitrate Lyase, Arginine Kinase, Protein 14-3-3 and HSP90 genes were selected, isolated and subcloned into the binary vector pK7GWIWG2 (I) for Nicotiana tabacum transformation via Agrobacterium tumefaciens. Furthermore, the expression of these genes was quantified and compared between the different stages egg, juvenile 2 and female, using real-time PCR (qPCR). The qPCR results showed that the genes Isocitrate Lyase, 14-3-3 protein were down-regulated and Arginine Kinase up-regulated in J2 when compared with the transcripts of egg. In female, the Isocitrate Lyase gene was up-regulated while Arginine Kinase and HSP90 were down-regulated when compared with transcripts of egg. Currently transgenic tobacco plants are being transformed and, after obtaining a homozygous line, these plants will be challenged with nematodes and the effects on their life cycle will be assessed.
