Description: The establishment of a germplasm bank using somatic cells is an important step prior to the use of genetic material in the bovine cloning process. Considering that each freezing curve temperature has specific implications for cell viability and functionality, the objective of this study was to evaluate the effect of three freezing protocols for fibroblasts from a high-genetic-merit dairy Sindi bull intended for cloning: 1. Pre-freezing at -80°C for 24 hours followed by immersion in liquid nitrogen (LN2); 2. Pre-freezing at -20°C for 24 hours followed by immersion in LN2; 3. Cooling to 5°C for 1 hour, gradual freezing in LN2 vapor for 20 minutes, and storage in LN2.
